Mechanism of action

Selectively activates mitophagy — the autophagy of damaged mitochondria — through PINK1/Parkin signaling. The mechanism is similar in effect to caloric restriction or exercise's mitochondrial benefits. Only about 40% of humans produce urolithin A from dietary precursors due to gut microbiome variation; direct supplementation bypasses this issue.

History

Urolithin A was identified as the bioactive gut microbiome metabolite of pomegranate ellagitannins in the 2000s. Amazentis (a Swiss company) developed a manufacturing process for synthetic urolithin A and launched Mitopure in 2020. Andreux 2019 demonstrated muscle function improvement in older adults. The compound represents a new class of mitochondrial-targeting supplements with stronger evidence than pomegranate extract for the mitochondrial effects.
