Mechanism of action

Punicalagins and other ellagitannins are metabolised by gut bacteria to urolithin A and related compounds. Urolithin A is the proximal mediator of mitochondrial benefit — Mitopure (synthetic urolithin A) is a purified version. Direct antioxidant activity. Endothelial function support. Anti-inflammatory through NF-κB suppression.

History

Pomegranate has been used in Middle Eastern, Mediterranean, and Indian traditional medicine for millennia. Modern interest accelerated with the identification of urolithin A as the bioactive metabolite. Multiple cardiovascular trials support arterial health benefits. The urolithin A direct supplementation (Mitopure from Amazentis) emerged in the 2020s.
