While PRL-8-53 has been around since the mid-70s, research is relatively limited, and there’s still a lot we’ve yet to discover about this drug. But, there’s also a lot that we already know. What is PRL-8-53?

PRL-8-53’s name may be a bit obtuse, but it’s certainly easier to say than the drugs actual name; methyl 3-(2-(benzyl (methyl) amino) ethyl) benzoate. This supplement was discovered by professor Nikolaus Hensl at Creighton University in the early ‘70s, and it was first patented in 1974.

PRL-8-53 is derived from benzoic acid, which can be found in many of our favorite foods, such as dairy products, berries, and some plants.

While there isn’t a large body of research to support the use of PRL-8-53, neurohackers have found it to be a powerful nootropic supplement that’s capable of improving memory function as well as cognition.

How does PRL-8-53 work?

When it comes to how PRL-8-53 works, that’s the million-dollar question. While preliminary research conducted in the ‘70s was promising, this supplement has largely taken a back burner to other new substances which have received more comprehensive testing. Since there have only been a few studies on the topic, we only have a preliminary understanding of how PRL-8-53 works.

Researchers believe that PRL-8-53 displays cholinergic properties and that it increases acetylcholine activity, which is a neurotransmitter that plays a critical role in memory and cognition. It looks like this supplement may also increase dopamine levels in the brain, which are connected to our memory function, motivation, and fatigue levels.

PRL-8-53 also seems to inhibit serotonin production, which may increase feelings of anxiety, depression, or insomnia. While heightened levels of dopamine combined with lowered levels of serotonin may lead to impulsive aggressive disorders, human trials did not reveal such symptoms or side effects.

PRL-8-53 benefits

The primary benefits of PRL-8-53 supplementation are improved memory and improved cognitive function.

The one human study of PRL-8-53 involved testing subjects based on their ability to correctly recall the order of 6-8 words in a word recollection test.

While those who scored the highest on the test in the control group didn’t seem to experience a vast improvement in their test scores on PRL-8-53, those from the control group who initially scored lower on the test showed a dramatic improvement in their abilities after PRL-8-53 supplementation.

Those who scored well in both the placebo group and the PRL-8-53 group did see a slight improvement in their scores after PRL-8-53 supplementation; it wasn’t deemed statistically significant.

The results were especially profound when looking at the improvements in test scores of individuals who were over the age of 30. This test suggests that those who have difficulty with memory retention stand to enjoy the most significant benefits of PRL-8-53, especially those over 30 years old.

A second experiment conducted by the drug’s creator, Dr. Nikolaus Hansl claimed that PRL-8-53 supplementation improved cognitive performance in test subjects as well, but the findings from that study are unpublished. Within that same study, Dr. Hansl also concluded that verbal proficiency and pattern analysis was also improved by PRL-8-53.

Of course, it’s difficult to conclude an unpublished study. But, if the thoughts of the nootropic community are of any consolation to you, then PRL-8-53 appears to be a powerful nootropic that indeed does offer powerful memory and cognitive benefits.

PRL-8-53 dosage

Like all of the other information about PRL-8-53, dosage information is quite limited. Most users take their cues from the one published study on PRL-8-53, which administered 5mg doses of the drug. After developing a better understanding of how they react to PRL-8-53, users often up their dosage to 10-20mg per day.

The patent information for this supplement states that the ideal dosage range per kilogram of body weight is .1-4mg. That’s an incredible wide dosage range. For example, someone who is 5’10 and weighs 80kg would have an ideal dosage range of 8-320mg per day, which is an incredibly wide variance in dosage.

While there’s no evidence that higher doses of PRL-8-53 are especially dangerous (the drug exhibited an incredibly high LD50 in animal tests, with a dose of 860mg/kg of bodyweight proving lethal) there’s also nothing to suggest a therapeutic dosage range beyond the 5-20mg range.

Ignoring the dosage information associated with the patent, we suggest getting started with a dosage of 5mg and adjusting your dosage up or down as needed, depending on the effect that is produced.

How to take PRL-8-53

In the published studies, Dr. Hansl’s team hinted that other administrations of PRL-8-53 might prove to be more effective, but only oral supplementation was studied. For that reason, we recommend sticking with an oral dose of this supplement.

PRL-8-53 is most typically available as a power, and it’s sold in 1-5-gram jars. Most users will weigh out their dosage and swallow it, add it to a drink, or package it inside of a capsule for easier consumption.

PRL-8-53 does seem promising. Many users in the Reddit nootropic community reported significant memory improvement with dosages in the 10-20mg range and were able to recall key themes, concepts, and figures with ease. They also had an easier time reiterating those concepts and themes verbally.

Some users have suggested that their reaction times when playing games were also improved, and they were more able to focus on the task at hand.

PRL-8-53 side effects

In human studies, PRL-8-53 was tolerated quite well, and the test subjects presented no side effects at all. In animal studies, high doses of PRL-8-53 appeared to depress motor function. Some users have noted that this supplement doesn’t interact well with antihistamines.
